چکیده

We report a comprehensive large-scale expression profiling analysis of mammalian male germ cells undergoing mitotic growth, meiosis, and gametogenesis by using high-density oligonucleotide microarrays and highly enriched cell populations. Among 11,955 rat loci investigated, 1268 were identified as differentially transcribed in germ cells at subsequent developmental stages compared with total testis, somatic Sertoli cells as well as brain and skeletal muscle controls. The loci were organized into four expression clusters that correspond to somatic, mitotic, meiotic, and postmeiotic cell types. This work provides information about expression patterns of approximately 200 genes known to be important during male germ cell development. Approximately 40 of those are included in a group of 121 transcripts for which we report germ cell expression and lack of transcription in three somatic control cell types. Moreover, we demonstrate the testicular expression and transcriptional induction in mitotic, meiotic, and/or postmeiotic germ cells of 293 as yet uncharacterized transcripts, some of which are likely to encode factors involved in spermatogenesis and fertility. This group also contains potential germ cell-specific targets for innovative contraceptives. A graphical display of the data is conveniently accessible through the GermOnline database at http://www.germonline.org.

Garlic (Allium sativum L.) fertility: transcriptome and proteome analyses provide insight into flower and pollen development

Commercial cultivars of garlic, a popular condiment, are sterile, making genetic studies and breeding of this plant challenging. However, recent fertility restoration has enabled advanced physiological and genetic research and hybridization in this important crop. Morphophysiological studies, combined with transcriptome and proteome analyses and quantitative PCR validation, enabled the identifi...

Systematic enrichment analysis of microRNA expression profiling studies in endometriosis

Objective(s): The purpose of this study was to conduct a meta-analysis on human microRNAs (miRNAs) expression data of endometriosis tissue profiles versus those of normal controls and to identify novel putative diagnostic markers. Materials andMethods: PubMed, Embase, Web of Science, Ovid Medline were used to search for endometriosis miRNA expression profiling studies of endometriosis. The miRN...

P-111: EGFR, ERK, MEK Genes Expression Level in Cumulus Cells of PCOS Women Compared with Healthy Women

Background Poly cystic ovarian syndrome (PCOS) is known as a common endocrine disorder in women at reproductive ages and may cause developmental abnormality in oocyte. ERK has found as a regulator protein of Gap junctions (GJ) function and the level of exchanges between two neighbors cells, for example oocyte and surrounding cumulus cells (CCs) in the mammalian ovary. Such exchange is essential...

I-51: The Role of the Transcription FactorGCNF in Germ Cell Differentiation and Reproductionin Mice

The germ cell nuclear factor (GCNF) is a member of the nuclear receptor super family of transcription factors. GCNF expression during gastrulation and neurulation is critical for normal embryogenesis in mice. GCNF represses expression of the POU domain transcription factor Oct4 during mouse post-implantation development in vivo. Oct4 is thus down-regulated during female gonadal development, whe...
